TOSOH ST AIA-PACK Intact PTH
Tosoh Bioscience has launched a test to measure intact PTH for the AIA-series.
Parathyroid hormone (PTH) is secreted by the parathyroid glands as a polypeptide containing 84 amino acids. PTH is secreted under the direct influence of a low calcium concentration in the blood. PTH acts to increase the concentration of calcium in the blood in three ways. It enhances the release of calcium in the bones, by activating the osteoclast. It enhances the tubular re-absorption of calcium in the kidneys and increasing the production of vitamin D, enhancing the absorption of calcium in the intestine.
PTH is rapidly metabolised in the liver and kidney to primarily biological active N-terminal and biological inactive C-terminal fragments with much longer half-lives.
Intact PTH is very suitable marker for:
- discovering abnormalities of the parathyroid gland like hypoparathyroism and primary hyperparathyroidism,
- judging success or failure of the removal of the malignant parathyroid gland,
- observing postoperative recovery.
Primary Hyperparathyroidism:
It is mostly an asymptomatic disease with a high prevalence in the population above 65 years old. According to different sources the annual incidence for males and females above 65 is respectively 1/1000 and 2-3/1000
Principle of the assay:
The ST AIA-PACK Intact PTH is a two-site immunoenzymometric assay, which is performed entirely in the AIA CUP. Intact PTH present in the test sample is bound simultaneously with antibody immobilised on magnetic beads and enzyme-labeled antibody. After 10 min. incubation the magnetic beads are washed to remove unbound materials. A fluorogenic substrate, 4-methylumbelliferyl phosphate, is added for enzyme-substrate reaction at 37C. The rate fluorescence measurement of converted 4-methylumbelliferone is directly proportional to the intact PTH concentration in the sample.
